Kualitas udara dalam ruangan kamar kos mahasiswa berisiko menurun akibat keterbatasan ventilasi alami dan tingginya aktivitas penghuni, khususnya merokok, yang dapat memicu akumulasi polutan berbahaya dan meningkatkan risiko kesehatan penghuni ruangan. Penelitian ini bertujuan mengevaluasi pengaruh kinerja ventilasi alami terhadap kualitas udara dalam kamar kos mahasiswa melalui analisis Air Changes per Hour (ACH), konsentrasi polutan utama (TVOC, HCHO, dan PM2.5), dinamika akumulasi polutan selama waktu paparan, serta penilaian kondisi udara terintegrasi menggunakan Indoor Air Quality Index (IAQI). Penelitian dilakukan pada 40 kamar kos mahasiswa di Kecamatan Lowokwaru dengan ventilasi alami. Pengukuran konsentrasi TVOC, HCHO, dan PM₂.₅ dilakukan selama periode hunian 8 jam. Nilai Air Changes per Hour (ACH) dihitung dan diklasifikasikan berdasarkan standar ASHRAE, kemudian dianalisis secara deskriptif menggunakan pendekatan keseimbangan massa dan Indoor Air Quality Index (IAQI). Hasil menunjukkan peningkatan ACH berkaitan dengan penurunan konsentrasi rata-rata polutan gas, namun kurang efektif terhadap PM2.5. Kamar dengan aktivitas merokok memiliki konsentrasi dan variabilitas polutan lebih tinggi pada seluruh kategori ACH. Analisis temporal dan IAQI menunjukkan ventilasi efektif pada kondisi tanpa aktivitas merokok, tetapi tidak mampu mengendalikan akumulasi polutan pada kondisi merokok.
